We have cultivated a choice of plant cell strains: traditional and new
strains o f P. ginseng - IFRZH-1 and DAN - 25, strain VK - 39
Lerythrorhizon
and strain ME - 75
A.euchroma.
These cells were grown in callus and
suspension cultures. The growth medium composition and other growth
conditions were analogous to those described in the normative documents.
Acco rd ing to recomendations o f investiga to rs preceded prepared
penopolyurethane was brought into the cultivation flasks before sterilization:
as cube (with 0,5 mm edge) for suspension cultures and as round bearers
(with 15 mm in thick and diameter to be equal to flasks' one) for callus
cultures. At the close of the cell cultivation cycle the yield of the wet and dry
biomass, the content o f biologically active substances (ginsenosides on
cell biomass
P.ginseng
and shikonin derivatives in cell cultures
Lerythrorhizon
and
A. euchroma)
were determined.
It has been established that penopolyurethane influence on growth and
biosynthetic activity depends on the type and strain of cells, methods of
cultivation and otther factors not controlled. Penopolyurethane does not
immobilize cells in suspension, but shikonin has not been found in cultural
fluid, whilst without penopolyurethane the shikonin content in culture medium
is nearly equal to that in biomass. Contrary to the available information there
are no reasons to assume that penopolyurethane has essential influence on
the levels o f biomass and biologically active substances in cell cultures. As
it follows from the results penopolyurethane makes possible callus cultivation
w ithout agar with the same yields of biomass and biologically active
substances. In suspension cultures o f cells - producers o f shikonin
penopolyurethane may be used for elaboration of alternative method of
shikonin separation from the cultural fluid.
