The capability of T-DNA agrobacterium to insert into host genome
determines a possibility to use it as vector in genetics transformation of
plants. Two clones of
Lotus corniculatus
with high regeneration ability have
been isolated in culture
in vitro.
Tile genetic transformation was produced by
method of stemps segments cocultivation with overnight culture of
Agrobacterium tumefaciens
containing plasmid
pTiBo 542
with marcer genes
res is tan t to hygromycin and kanamycin. Chymeric gene o f
phosfinotricineacetyltranspherase (bar) containing in vector determines
resistant to herbicide fosphintricine (BASTA). Green shoots were obtained
on selecting medium with 50 mg/l Km. Presence of
nptll
-gene in transformed
plants was determined by PCR-analyse. Transgenics plants phenotype is
the same as control. Rowers were fertility and produced normal seeds. Four
clones transgenics plants are resistant to BASTA in greenhouse in dose 0,4
kg/ga.
