OPTIMIZATION OF CLONAL GRAPEVINEMICROPRAPAGATI ON

Meristem ic explants of small (0,17-0,25 mm) and extras- mall (0,075-
0,1 mm) sizes were used fo r healthening plants against phyloxera,
nematodes, fungi and bacterial pathogene infection. To increase their
regeneration activity the phase o f introduction was divided into two with,
alteration of hard and liguid culture medium, content o f 6-BAP at every phase
was defined and the possibility of low intensity EMI applying was investigated.
It was determined that influencing of EHR rays togetherw ith narrow stripped
lazers upon meristem, planted in hard culture medium, resulted in 2,5 folds
their regenerative ability. Ways o f optimization clonal micropropagation during
microculting
in vitro
plants were investigated: applying of EHR rays, vegetative
supplement o f fine-milled grapevine seeds, introducing o f 0,05 mg/l 6-BAP
into culture medium. Daily growth rate increases in 1,7-2,4 times under
EHR raysinfluencing, plant weight (mass) twicly, shoot length in 1,7, node
formation number 1,2-1,5 folds. Applying natural growth stimulators, o f fine
- m illed grapevine seeds into cultural medium, increases to 27,4% the
effectivness of clonal micropropagation. Introduction o f 0,5 mg/l 6 BAP into
culture medium increases microcutting more than twicly. Improving ways o f
plant adaptation permits to rise surviving ability of plants in the process of
laboratory adaptation up to 96,6-97,4%. The technology o f laying nursery
w ith healthied plant material of clonal micropropagation was developed fo r
stationary greenhouses as well as fo r covered ones and in tho opend soil.
Following this recommendation procides high surviving ability (80-90%), good
shoot development (117,6-184,1 cm) and maturing (109,1-138,7 cm) wich
0,5 cm in diametre, thats why in a year it is possible to lay nursary and next
year to begin preparing cuttings.
