very difficult to choose criteria. Using of selective systems enables to receive
resistant variants, it does not always correlated with synthesis secondary
metabolites. Visual markers can facilitate selection of productive clones. In
this respect
Thalictrum minus
is good object, as the quantity berberine,
correlates with color of cells and medium. To obtain stable cell lines to
many traits is complicated in connection w ith significant variation at
cubcultivation. Therefore it is important to know, when and how to estimate
clones in absence o f the selective factors.
For the decision o f this task primary selected on intensity o f color agar
medium seven clones were suspended and during 20 cubculture, were
analyzed the berberine content on the medium by spectrophotometry method.
During eight cubculture the berberine contents, as a rule, does not
exceed its in the initial strain and change from passage to passage (coefficient
o f a variation from 50 up to 90%, in the control - 30 %). Berberine content in
medium from selected clones was stabilized after 1,5 years (decrease of
coefficient of a variation). One part of clones observed precise decrease the
contents alkaloid, in others - increase.
As a result it was obtained two clones 124 and 233the berberine content
o f them were up to 420 and 440 mg/l, that higher the control in 2,5 times.
