The ways o f prolonged grapevine storage

in vitro

bymeans o f growth

m inim ising test-tube plants were investigated: prolonged storage under

positive lowering temperatures, applying CCC, introducing mannitoi and

sorbitol, 6-BAP-into culture medium and natural growth inhibitors. Positive

lowering temperature (10,5-12,0°C) and lighthess (5 0 0 l) promotes growth

lim ita tion o f te st-tub lep lan ts and more prolonged cu ltiva tion w ithout

transplanting. New storage conditions o f collection grapevine genofond

in

vitro

w ithout transplanting during 12 months and more by means o f applying

specisi medium and t 4-5°C were developed in cooperation with the laboratory

o f criostorage o f T im iryazev’s Institute o f plant Physiology. Increasing

prolonged plant storage up to 4 months applying 60 mg/l sorbitol into culture

medium , up to 6 months applying I mg/l CCC were proved to be possible.

Introducing of 0,1 mg/l BAP into culture medium and increasing saccharase

content up to 6% influenced the dem inishing root length o f test-tube plants

more then 6 tim es and shoot lihgth 3,5 times. This achieved growth

m inim isation can be used fo r more prolonged storage o f grapevine genetic

fund. Applying milled grapevine seeds in the amount o f 1,0-2,0% o f medium

volume gives such level of, natural inhibitors (abscisin acid, phenols, and so

on) at which root formation decreases in 1,7 times and root growth in 3,6,

shoot length in 3,5-5,7, their mass deminishes in 3-5 times node number on

the shoot more then in four times. As a sequence the periods between plant

fransplantation into fresh culture medium increases in 3-5 times that can be

done in 5-8 months instead o f 1,5-2 month. It rises the economic effectiveness

o f storage, as labour expanses decreases and growth inhibition of test-tube

plants is reached w ithout additional wastes o f labour and time.
