OBTAININGOF NEW PSTV-FREE LINES OF POTATO PLANTS OF
RUSSIAN CULTIVARS BASING ONMODIFIED TECHNOLOGY FOR
DIAGNOSTICS AND VIRUS ERADIATION

Eradiation o f viruses through tradional tip meristem method does not
garantie eradiation o f PSTV. S tock plants in gene banks obtained from tip
meristem are,as a rule,virus-free, but they are not obviously PSTV-free, as it
can survive after traditional chemical and thermotherapy, inactivating viruses.
To determ in wether it is possible to eradiate PSTV effect of low temperatures
was studied both with and w ithout nucleases. The effect was studied upon
tip meristem culture, tube plants and tubers. PSTV detection in regenerant
plants was perfermed by three methods:1 .indicative plants; 2. lectrophoresis;
3 .m o lecu la r hybridization. Our technology o f eradicaton included several
new stages: 1.Tuber seedlings selected fo r obtaining o f healthy clones were
tested fo r PSTV by available methods; 2.P!ants obtained from indexes
characterized as "healthy" by the first PSTV test were tested again after
PSTV provocation (illumination and high temperature(30-35 degrees C); 3.Tip
meristem , obtained from plants, characterized as PSTV-free after first two
tests, was cultivated
in vitro
w ith application of several therapy elements
(low 2°C) temperature, nuclease addition into the medium); 4.P lants being
grown
in vitro,
in glass house and in field were tested three times by three
independent methods. As a result of PSTV tests (including those performed
by Agd ia company, USA) it was shown that new lines o f cultivars Udacha,
Lukjanovsky, Newsky, Pushkinets, Lugowskoj and others are PSTV-free.
The development o f a new, PSTV-free collection o f genotypes has been
started fo r the cultivars, included into State Register of Russian Federation.
