APPLICATION OF BIOLOGICALLY EFFECTIVE SUBSTANCES TO THE TIP
MERISTEM CULTUREWHILE OBTAININGVIRUS-FREEPOTATO

To increase effectivity of obtaining o f virus-free potato, the tip meristem
method was coupled with chemiotherapy. The following substances were
applied: 1.bacterial endonuclease (100000-200000 units per a liter); 2. bacterial
ribonuclease (100000-200000 units per a liter); 3. deoxyhexahydro-1,3,5-
tryazin (neo-DGT), (10 0 mg/l); 4. allylthioureidobenzoic acid (O-ATB), (1,0-
2,0 mg/l); 5. orthophenylureidobenzoicacid (O-PTB), (1,0-2,0mg/l); 6. ridostin
(dsRNA from S.cereviseae killer strain), (4-8 mg/l); 7.riphastin (dsRNA of
fage F6), (4-8 mg/l); 8. PE-26 (substance obtained from arachidonic acid),
(dissolved 1:1000000); 9.34-S (a modified form o f emystim with arachidonic
acid),(dissolved 1:100000); 10.emystim SHK (form34), ( dissolved 1:100000).
Investigations performed have shown that application o f biologically
effective virus-inhibiting substance resulted in lower concentration of viruses
in tube plants, but their effect was virus-specefic. For example, 34-S inhibited
X,S and M potato viruses, the effect was particularly noticeable when
concentration of the substance in the medium was 10-6. Whem emystim
was added into the mediumthe concentration was 27% less fo r PVX, 10%
less fo r PVS and 13% less fo r PVM in comparison to control plants. For
PE-26 decreasing of PVX, PVS and PVM concentration was 28%, 35% and
32% accordindly. When form 34 was present in the medium PVX concentration
was less by 45%, but PVS and PVM were equal to that of control plants.
An tivira l substance riphastin reduced PVX and PVS content in plants by
28% fo r the first passage and by 30% fo r the second passage. Ridostin
application resulted in less concentration of PVX, PVS, PVM and PVL. The
difference was 10-32% depending on the cultivar. Thus, the data obtained
show that application of biologically effective substances are very prospective
fo r obtaining of virus-free potato. The greatest effect was observed for bacterial
endonuclease and ribonuclease.
