THE USE OF

IN VITRO

CULTURE METHOD FOR PROPAGATION OF

MEDIGINALPLANTS

Gontinuons destruction and slow restoration of populations of rare plant

species - sources o f valuable medicinal raw material show that measures of

protection and regeneration o f their gene pool are very actual. In addition to

the use of traditional methods such as, the establishment of preserves and

sanctuaries the cultivation o f these plants

in vitro

is necessary fo r solution

o f the problem.

Rhodiola iremelica Boriss., R. rosea L , Valeriana officinalis

L , Paeonia anomala L.

belong to rare species recordet in "Red book o f

Bashkortostan Republic" and "Red book o f USSR". Thickets of

Polemonium

coeruleum L .

are also not numerous because o f intensive store. Models of

micropropagation used in the investigations are proliferation of axillary buds,

producing o f callus tissue withe the following induction o f organogenesis.

The technology o f clonal micropropagation o f

R.rosea

and

R.iremelica

including explantation of initial tissue, properly micropropagation, rooting of

propagated shoots, propagation of regenerated plantlets for planting in opened

substiatum , introduction o f plantlets in nature, primary chem ical and

ensymatic estimation of their raw material is developed in laboratory. Selekted

optimal conditions fo r cultivation

in vitro

of both

Rhodiola

species are based

on study o f in trabud deve lopm en t, ana tom ica l and m o rphologica l

construention of buds, their hormonal status as well as on selectio of

com position and consistence o f nu trien t med ia and so il m ixtures.

Comparative estimation of raw material o f wild, untroduced in culture and

in

vitro

regenerated plants is conductifed. Primary pharmacological study of

tinctures from raw material of regenerated plantlets shows the possibility of

their use for preparation o f medecinal compounds of stimulating action. Mass

production o f

R.iremelica

piantlets proceeds with the aim o f reintroduction of

species in natural locations. Technology of micropropagation of

P.coeruleum

L

is being developed using the avobe scheme. High coefficient of multiplication

up to 160 shoot per one exsplant obtained. Experimentally-commercial plot

is beineg created nohich may be serve as a base fo rdevelopment of industrial

technology o f raising o f planting material. Sterile culture of

V.officinalis

and

P. anomala

is obtained. The conditions for different steps o f

in vitro

cultivation

o f these species are being optim ized now.
