PRESENCEOFHERBICIDESINACULTUREMEDIUM

Selection of somatoclonal variants on selective culture medium is a

perspective method for creation of the new forms of plants, in particular,

forms which are stable to various abiotic factors of external environment.

However, the use of this method for some agricultural cultures is limited by

the insufficient studies of influence of the selective agents on the processes

of regeneration.

The purpose of this research was to determine an opportunity of selection

of the forms which are more stable to herbicides. The sensitivity of isolated

shoots and proliferous cultures of seven sorts of strawberry (Gora Everest,

Dukat, Zenga-Zengana, Geneva, Redgontlit, Tribute, Tristar,) cultured

in vitro

with the presence of two types herbicides in a culture medium was studied.

The inclusion of Symazin (the range of concentration 2x10 -5- 2x10-

4M) has re-sulted in the inhibition of explants growth and development.

The 10-3M concentration caused the symptoms o f chlorosis up to

comp le te loss o f the coloration w ith the subsequent destruction o f

explants o f all sorts. Among tested samples sort Geneva was the most

s e n s itiv e to the p resence o f S ym a z in in cu ltu re m ed ium . The

concentration o f 10-4M had appeared to be lethal fo r its explants. The

explan ts o f othe r sorts were more to leran t and the selective threshold

o f concentration fo r this herbicid in condition

in vitro

was at a level 2 x

10-5M. Studying the effect o f Roundup (gliphosat) inclusion in a culture

medium it was revealed that the three highest concentrations (10-4; 2 x

10-4; 10-3M) resulted in suppression o f explants growth and development

at the first stages o f culturing, the leaves became brown and by the end

o f the first month of culturing they perished. The concentration o f 10-5М

and 2 x 10-5M should be considered as the threshold and recommended

as se le c tive fo r the given preparation. For confirmation of increased

s e n s itiv ity o f selected explants they were recultivated on a culture

medium , containing sublethal concentrations of appropriate herbicides.

The to le ran t plants of strawberry selected at subsequent culturing are

being tested in fie ld conditions for distinguishing o f their degree o f

tolerance to herbicides.
