Successful application o f methods of tissue and organs culture in sugar
beet breeding makes the question o f revealing and using of molecular genetic
markers in biotechnological works actual.
Isozyme analysis o f donor genotypes and obtained regenerants was
carried out w ith the aim to determ ine the opportunities to use isozymes as
genetic criteria when culturing un-fertilized ovules and petioles o f sugar beet
in vitro.
It's found, that donor genotypes, heterozygous in isozyme markers,
formed the most haploid plants (64%) o f the total microclones obtained.
Isozyme control showed their 100% heterozygosity in loci Me-1, ldh-1, Gdh-
1
.
Initial plants and regenerants, obtained from them, when culturing
petioles on selec-tive media, modeling abiotic stresses, caused by salinization
and herbicide chlorsulphuron, were assessed in 6 isozyme loci (Me-1, Mdh-
1, Mdh-2, ldh-1, ldh-2, Pgd-1, Gdh-2).
Identity o f isozyme spectra of control and tested plants was shown.
There was revealed the modification in malic-enzyme of regenerants,
obtained from callus as additional isoforms with less electrophoretic mobility
o f the frequency o f 10%, that, probably, is a result o f mutations, affecting
genome.
Thus, isozymes can be widely used as molecular genetic markers in
different biotech-nological investigations.
