IDENTIFICATION OFTHE NEWGENESWITH THEAС/DS ENHANCER-

TRAP SYSTEM

Using the Ac/Ds maize transposable element and bacterial and plant

marker genes a binary vectors were made and used to identify the plants

where the excision and the reinsertion of the transposon somewhere in the

genome occurred. The presence of the b-glucoronidase gene (GUS) in the

vectors allows the detection o f the transpositions .After the transformation

o f the Arabidopsis plants with these vectors a system of enhancer-trap lines

was obtained that allowed marking and identification o f the genes with the

tissue specific expression. Using the new method TAIL PGR allow us to

determ ine the transposon flanking sequences and as the result to get the

clones of several new genes and among them a SSR16 gene.The transposon

insertion into the gene sequence caused the lethal mutation, arresting

embryoid development at the globular stage. Earlier the possibility o f the

maize Ac element transpositions in the carrot, potato and tomato genomes

was shown that make it possible to use the same enhancer-trap system for

the identification o f the new genes.
