Initial material was mature and immature embryos o f 8 genotypes (
W W , 81S-10,91S-15, Enita, Moskovskaya-35, Saratovskaya-29, fortune,
Gaterin temps). To obtain the primary callus, isolated embryos were cultivated
in modified media MS, containing inosit 100 mg/l, asparagin 120 mg/l,
ascorbic acid 20 mg/l, sucrose 2%, agar 0,7% and also 2,4D 2mg/l. As a
selective factor utilized 14th day cultural filtrate (CF) of fungi
Septoria nodorum,
which was obtained by cultivating fungi in liquid nutrient media MS in conical
flask in shaker. Cellular selection were carried out in aggregated media,
containing CF in concentrations 5 ,10 ,1 5 , 20%. There was two scheme of
selection: 1 )4 passages in selective media, 1 passage without selective
factor, 4 passages in selective media. 2) cultivation o f callus in selective
media w ith increasing concentrations of CF from 5-20% w ith every
subcultivation (4 passages), 1 passage without selective factor and repeated
cultivation of callus in selective media with increasing concentration of CF
from 5-20%.
The obtained results allowed to exposure some phenomenon which
happened with callus independently from utilizing selection schemes. In the
first passage o f selection about 85% o f callus were formed with medium
density, bright yellow coloured, but in the further subcultivation 50% callus
were formed with brown coloure without any sign o f morphogenesis. Further
cultivation o f callus in a non selective media causes to rehabilitate the
proliferation activity. Repeated utilization o f selective factor helps to select
callus which are resistant to CF o f S. nodorum after 8 passages in selective
media. Regenerates were obtained from resistant callus o f mature embryos
genotype Enita, and callus o f immature embryos genotype-81 S-10.
