The capacities fo r regeneration, callus formation, and organogenesis

w ere studied in tobacco (

Nicotianatabacum L .)

plants, both wild-type (control)

and transform ed w ith the active ipt gene o f agrobacterium . Control plants

w ere capable o f form ing calli only during 7 weeks, transgenic - up to 10

weeks. High frequencies o f callus formation were observed in control explants

when various concentrations o f 2,4-D (2 and 1 mg/l) and kinetin (0,2 and 0,5-

1 m g/l, respectively) were added to the m edium ; in transgenic explants -

only when 1 mg/l 2,4-D and 0,1 mg/l kinetin were added. W e observed

auxotrophic form ation o f the primary ipt callus tissue. The morphogenic

potential of ipt callus proliferated on hormone-free medium could be improved

by adding kinetin. Regenerated shoots form ed by ipt explants in the light

varied in their morphology: easily rooting, normally developing plantlets;

rosette plantlets lacking roots and num erous m icroshoots. In darkness

shootlike and nonviable abnormal structures were form ed. Morphological

traits o f ipt callus are probably conditioned by their specific hormonal system.

Transgenic plants have been recieved from prof. E.S.Piruzian (Institute

o f M olecular Genetics RAS, Moscow).
