Ti-plasmids and leaf discs method with some modifications

were used. As a target gene served human

α

-interferon gene or a dsRNA

coding sequence. As a selective marker these sequences carried NPTII

gene. The target gene in potato genotype was detected by NPT-test at first,

then by Southern blot-hybridization or PCR. Evaluation of transgenic plant

resistance to PVX was performed by determination of PBX concentration in

plants grown upon sand, saturated with feeding medium and infected

artificially. Measuring of PVX contents performed by ELIZA-method in 6-30

days after infection. Data obtained show the ability of interferon or dsRNA,

known to be an inductorfor interferon-like plants proteins, to modify resistance

(sensitivity) of potato plants to PVX infection within a wide range. The virus

concentration in plants varied from several times increasing comparing to

control plants up to a level, close to a partial resistance. In some cases the

contents of a viral antigens appeared to be lower then ELIZA-method

sensitivity. Resistance of several transgenic lines varied from one experiment

to another. Thus the lines obtained can be conditionally divided into 3 groups:

1. Plants revealing some degree of resistance and supporting virus

multiplication at lower level in comparison to control plants o f initial cultivar;

2. Plants more sensitive to virus in comparison to initial ones;

3. Plants with a resistance which varies from experiment to experiment

within a rather wide range.

Data obtained could be, in our opinion, connected with multieffect of

interferon upon plant regulation systems and, privately, upon the ir

immunological status, which can, as for 3-d group, became unstable.
