THE EXPRESSION INSTABILITY OF TRANSFERRED GENES IN
TRANSGENIC PLANTS (
GENETICMODELS)

transgenic plants is presented as the undesired phenomenon at plant genome
modification by gene engineering methods. Some genetic models for study
o f molecular mechanisms and reasons of foreign genes inactivations are
created in the Institute of Cytology and Genetics (Novosibirsk).
The line To-5
(Nicotiana tabacum, cv.Gutersleben
) includes three alleles
of marker nptlI gene (resistance to kanamycin) revealed at analysis of selfing
progenies of this line. The genetic analysis of nptll gene is conducted. The
recessive character of allele nature supervising inactivation of nptll gene is
established. The cases of epigenetic changes of transgenes inactivation/
reactivation are revealed. The obtained results are tested on line 6/15
(N.tabacum, SR1). The series of transgenic plants (
N.tabacum, SR1),
simulated instability of transferred genes expression with duplications (direct
and inverted) on uidA gene is created. The increase of nptll gene expression
instability more than at 50 % plants is demonstrated. The line Nu2/1 presents
the unique model, showing mosaic nature of instability of nptll gene
expression on the level of somatic tissue. It was shown, that mosaic nature
of nptll gene is inherited in the progenies from selfing and from crossing with
untransgenic tobacco plants.
