COMPARINGGROWTH PARAMETERS OF STEVIA CELL SUSPENSION

WHEN CULITVATING IN THE FLASKSANDIN THE FERMENTER

To reveal the possibility o f stevia cells cultivation in the fermenter we

investigated several growth characteristics of earlier obtained suspension

when cultivating in the air-lift fermenter and in the flasks on the shaker.

Stevia cells were grown under batch cultivation at 26°C. We used basic MS

medium supplemented with NAA (1 mg/l) and BAP (0.5 mg/l). Cell number,

fresh and dry biomass and cell viability were detected within the cultivation

cycle.

Growth curves were similar for both methods of cultivation. The highest

possible specific rate of the growth was 0.22 d a y1for flasks and it was 0.25

d a y 1fo r the fermenter. The minimum periods of biomass doubling were 3.2

day and 2.8 day accordingly. Dayly biomass yield in the fermenter was 0.51

g/l, bat this index in shaking flasks conditions was 0.49 g/l. Sucrose utilization

officiency o f the first method was 0.31 and that of the second one was 0.34.

Thus, we have demonstrated the possibility to cultivate stevia cell

suspension in fermenter. The main suspension growth characteristics when

growing in the air-lift fermenter demonstrated a slight increase in comparison

with the same ones in shaking flasks conditions.
