ab53S0@mail.sitek.netStudies the capacity of the isolated wheat leaf explants to form
morphogenic callus depend up on the nutrient media. The work was carried
out with 5 genotypes (Moskovskaya-35, Fortuna, Enita, WW, Gaterin temps).
As the primary explant we used the basal part of the flag !eaf(in a size of 3-
5 mm) from sterilized 7th day germinates. Cultivation of leaf segments were
canted out in modified media MS, containing inositol 100 mg/l, asparagine
120 mg/l, ascorbic acid 20 mg/l, sucrose 2% and agar 0,7%. Studied the
influence of 2,4-D 2 and 3 mg/l for the callusogenesis and the combination
of 2,4-D 2 mg/l with zeatin 0,5 mg/l and 2 mg/l, and also NAA 0,5 mg/l and
BAP 0,5 mg/l for morphogenesis.
Established that the concentration of 2,4-D affected the capacity of leaf
explants to form callus. For the genotypes Moskovskaya-35 and Fortuna
optimum media was which contains 2,4-D 3 mg/l, for Enita, WW, and Gaterin
temps - 2 mg/l. In those conditions formed hard, green colored callus. By
transplanting the callus to the media fo r regeneration the best results were
obtained from the media which contains 2,4-D 2 mg/l and zeatin 0,5 mg/l. In
variants NAA with BAP more than 50% shows rhizogenesis. In this condition
regenerates were obtained only for Gaterin temps.
