One of alternative ways of getting regenerants from isolated tissue,

which can be transformed by different ways is the regeneration of shoots

from plants meristem. Apical meristem were dissected isolated from 7 day

s te rile sh oo ts o f co tton v a rie ty C -6524 , re fe rrin g to spec ie s

Gossypium.hirsutum L. Fo rthe cultivation of isolated meristems mediums

were used, the basis of which was composed of the salts MS, vitamins B5

according to Gamborg, agar 0,7%, glucose 3%. Phytohormones: 2,4-D,

IAA, BAP, kinetin in concentration 0,05 and 0,1 mg/l and NAA -2,0 and 5,0

mg/l in different combinations were added to the medium fo r the growth

induction and development of meristems. On medium, containing IAA in

combination with kinetin or with BAP, cotton meristems at first started to

grow, then their growth gradually slowed down, forming shoots gradually

became black and perished. 2,4-D in combination with both cytokinins has

stimulated the formation of callus tissue. At planting of meristems on the

medium, containing NAA - 2 mg/l the main part of isolated meristems started

to grow and the formation of shoots has begun, inconsiderable part of

meristem has started to form callus tissue. The most optimal combination

o f hormones fo r cultivation of isolated cotton meristem were NAA - 5 mg/l

with kinetin and BAP, moreoverthe latter combination stipulated for maximal

shoot growth. A t more prolonged cultivation on this medium shoots has

started to form roots.
