Optimization of conditions for getting of standard regenerants in tissue

culture cells and protoplasts is one of the important moment for experiments

fo r genetic modification of plants by gene and cell engineering methods.

The problem of the present work was the study of morphogenesis and

regeneration abilities in tissue culture Triticum aestivum of local varieties:

Intensiv, Trecum-106, Tez-pisharand collection lines of UzNIlR. For callus

induction immature embryos after 10-17 days pollination were taken. The

length of the embryos were 0,5-2,2 mm. Cultivation was conducted on modified

medium MS with the addition of vitamins according to Gamborg’s B5, agar

8 g/l, and 2,4-D (2-5 mg/l). The data fo r induced hcallus quantities, the

presence and quantities of morphogenetic zones and regenerated plants of

all studied genotypes were obtained.

Not identical ability to callusogenesis and plant regeneration o f different

genotypes was revealed. Frequency of forming callus clones and regeneration

clones on the number of initial explants varied from 21,5-100% and from

10,1% to 98% accordingly of different genotypes. It was noticed that at

concentration of 2,4-D - 2 mg/l the differentiation and morphogenesis process

go faster, the number of regenerants to the number of initial callus clones is

higher.
