The capacity o f callusogenesis, morphogenesis and embryogenesis
has been investigated for four prospective varieties of clover: VIC-7, Mars,
Rannij-2 and Arlington. The hypocotyl, cotyledon's leaf and leaf-stalk explants
were excised from plants grown aseptically. We used Gamborg B5, MS and
LS media with some modifications. Callus cultures were obtained from all
used varieties. All received callus cultures had morphogenic capacity and
preserved this property during subculturing.
LS medium with picloram (0,06 mg/l) and BA (0,1 mg/l) has been used
for morphogenesis. Varieties VIC-7, Rannij-2 have maximal capacity of
morphogenesis.
LS medium with 2,4-D (0,01 mg/l) and adenine (2,0 mg/l) has been used
for embryoids formation. For embryoids development the LS medium with
picloram (0,02 g/l) and BA (0,2 mg/l) was used. Regenerants have been
obtained from embryoids. NAA (0,2 mg/l) was used for root formation.
Ten regenerants of varieties Rannij-2 have been used for callus formation.
Callus cultures were obtained from all regenerants. Fertile seeds have
been obtained from some regenerants of variety Rannij-2.
