The electrophoretic spectrum and a-am ylases activity were studied at
various stages o f callus tissues and dihaploid lines morphogenesis, obtained
in the culture o f isolated wheat microspores
in vitro.
For studies o f the callus
tissues amylase activity the morphogenous and non-morphogenous lines of
haploid callus were used in experiments, identified by their cytoembryological
and morphophysiological features, obtained on the modified medium N6.
On the MS medium (2 mg/l 2.4-D ), as term s o f cultivation increased, the
a ctivity a-am ylase also increased "by stages" reaching its optimum value
on the 30th day o f cultivation.
On the MS medium w ith 0.5 IAA the optimum a-am ylases activity is
seen on the 15-20th day o f cultivation and later becomes lower. The reason
fo r such change in a-am ylase activity may be not only the difference of
conditions and terms o f cultivation, but also dissim ilar levels of development
and differentiation o f haploid callus ceils o f wheat.
In the research o f protein polymorphism, as well as o f a number of
enzymes, including a-am ylase, fo r dihaploid lines o f wheat under study
have differences o f in th is enzyme activity, whereas the electrophoretic
spectrum had no significant differences.
Thus, it was found that the degree o f heterogeneity o f electrophoretic
spectrum and a-am ylases activity is one o f the important factors at the
initial stages o f morphogenesis process and plants regeneration in the culture
o f isolated wheat anthers and microspores.
The discussion concerns the possibility to use the enzymes activity
on the example o f a-am ylases, to reveal the peculiarities o f dihaploid lines
o f wheat, obtained in the m icrospores culture and as biochem ical markers
fo r morphogenesis in cells culture
in vitro.
