In this w o rk a strictly photoperiodic line o f
Solanum tuberosum
, ssp.
andigena
forming
tubers only in short days w as studied. Recently it w as found that a night break (NB) fully
prevented tuber form ation o f this clone in
in vivo
experiments. T he aim o f the present study
w as to investigate w hether the N B reaction can be also reproduced in the course o f the
regulation o f tuber form ation in
in vitro
system w ith heterotrophic type o f nutrition.
Plants o f
Solanum tuberosum
, ssp.
andigena
w ere cultured
in vitro
for 1,5-2 months on
M S m edium supplem ented w ith 60 mg l-1 inositol , 0.5 m g l-1 thiamine, 0.5 m g l-1
pyridoxine, 6% sucrose and 0.8% agar. Three different photoperiodic treatm ents w ere
studied: a) short days (SD ) (14 h darkness, 10 h light); b) long days (LD ) (continuous light);
and c) night break (NB ) (as SD, but the dark period w as interrupted in the m iddle by 1 h
w hite light irradiation).
P hotoperiod influenced the m orphogenesis and the pattern o f grow th o f underground and
aboveground plant organs. T ubers w ere form ed only in SD variant and w ere absent in LD
and N B plants. Plants grow n in N B displayed many grow th characteristics intermediate
betw een SD and LD . Com parison o f the experim ents
in vivo
and
in vitro
show ed the
sim ilarity o f N B effect on tuber form ation and grow th pattern o f
Solanum tuberosum
, ssp.
andigena
plants.
T he presented resultes enable us to say that the developted
m odel for the
m orphogenesis o f potato plants
in vitro
may be used for the investigation o f regulatory
photoperiodic reaction in tuber formation.
