Using in selection system of triazol flingicides causes break of normal
sterol biosynthesis in plants by blocking of cylochrome P450-depent 14 a-
demethilation, In this work we used propiconazol us a seleclive agent which
inhibited clon formation of
S.tiberosum
cv.Tawa in 20 mkM concentration on
Murashige-Skoog agar medium. In selective conditions the treatment of cell
suspension by N-nitrozo-N-methilurea in I mM/h and 5 mM/h doses increased
the effect of the don formation. We yielded more than 800 clones in the
series of trials on different treatments of mutagen in 0-passage on selective
medium. There was selected 9 cell lines after 8 cultivation cycles on selective
medium. To verify the stability of resistance, the clones were cultivated 2
cycles
on non
selective medium. Then they were returned to selective
conditions again. The 5 from 9 cell lines maintained the capacity to grow in
se led ive condil ions. The analysis of dynamics o f sitosterol, stigmasterol
and campesterol content was carried out fo r callus tissue of resistant cell
lines and control ones. The level of campesterol content increased in more
than 28 times comparing to control one was observed in the presence o f
propiconazol in one cell line. After three non seledive passages the levels o f
sterol content were not different from control levels in this line.The level o f
sitosterol content o f the other cell lines increased in more than 50 times in
these conditions. Additional three non selective passages led to stabilisation
o f sterol content levels. In our opinion, the possible mode of resistance o f
the potato tissue culture to propiconazol is superproduction o f C 14 a -
demethilase.
