HMW glutenin composition is used for pre-selection in

early breeding generations (F3, F4). A higher total amount of

analyses is advisable. For classical electrophoresis from one

grain is necessary to analyse at least 5 grains per progeny. To

increase the labour capacity in the breeding process we use

mixed samples of

20-100

grains for electrophoretic analyses.

To verify this method we analysed artificially mixed samples

of two wheat varieties with different glutenin subunits and

bread making quality. The five percent share of cultivar can

be detected in a mixed sample on PAGE electrophoreograms

of HMW subunits of glutenins. The colour intensity of protein

bands enabled to detect the encreasing percentage of cultivar

with different HMW subunits in mixtures. The codominant

heritability enabled to detect parental or recombinant gametes,

homozygous or segregating progenies in HMW glutenin

subunits in F

plant progenies. In the breeding process in

later generations we analyse only selected lines which are not

homozygous in HMW glutenins. (Kubanek, Hanisova, Horcicka -

1999).

Mixograph is an efficient, simple and fast tool in wheat

breeding programme. In our programme we used mixograph

analyses since 1993 year. 35g, lOg, 5g and 2g versions were

developed. We are using lOg version. Different mixogram

parameters or indexes were used for evaluations (Martinant
