Long fruit stalk, fruit dense pulp, firm skin, high content of carotenoids-

not all the criteria, which breeder should take into account while the creation

o f new sea-buckthorn cu ltiva rs w ith application as trad itiona l and

biotechnological methods takes place.

Cultivation of callus tissue, with the following initiation of organogenesis,

should leads to changed plant obtaining. Meristem tissue (0,2-1,5 mm) was

used as an explant for callusogenesis initiation. It was placed on nutrient

media with different content o f mineral salts, vitamins, hormones and with

different physical conditions (full darkness, 16 hours light/8 hours darkness

photoperiod, different intensity of illumination and different temperature) used.

Organogenous callus tissue was obtained when the explant was replaced

on modified WSM ( Sviridenko-Burdasov) medium with hormones (in ratio:

BAP 0,5: IB A 1 ,0 :2,4-D 1,0) added and in the full darkness. To initiate the

organogenesis it is necessary to use the photoperiod 16 hours Iight/8 hours

darkness at 25 °C. The hormone ratio in WSM medium should be the following:

BAP 1 0 : IBA 1,0 :G A 0,5.

So rooted plantlets of sea-buckthorn transferred to soil substrate were

obtained.
