Combination of remote hybridization and

in vitro

culture enable the

possibility to overcome interspecific incompatibility. Interspecific hybridization

makes it possible to introduce valuable characters of wild species of

buckwheat into cultivated buckwheat. On the ground of caryological and

biochemical features genetic affinity of some buckwheat species was

established that helped to define the outline o f crossings. As mother

component of hybridization served cultivar Astra and Selenotsvetkovaya,

Homostylnaya and Belorusky Donor (Fagopyrum esculentum) forms, as

pollen donor F. tataricum var. rotundatum (k-17) and F. tataricum var.

tuberculatum (k-21) were used.

Application of growth regulators significantly stimulated pollen tube

growth in styles and fertilization in buckwheat interspecific crossings.

Conditions of hybrid embryos growth and age (12-14 days) were defined.

From callus embryo tissues multiple shoots were induced on MS basal

medium with the addition of phytohormones. Optimal composition of nutrient

media for further development of hybrid buckwheat plants and for their

microclonal propagation is developed. Addition of 1,2 mg/l NAA stimulated

root formation and 3 mg/l BA shoot development in hybrid plants. Hybrid

nature of developed plants was confirmed by isozyme analysis. While parents

differed by AHG and MOR loci hybrid plants carried both isozymes. The

majority of hybrids were fertile. Chromosome counts in squash preparations

showed tetraploid (amphidiploid) composition (4n=32) of most hybrid plants,

while parents were diploid (2n=16). Many morphological characters in hybrids

were intermediate between parents.
