ORGANO-AND EMBRYOGENESIS INNARROW-LEAVED OLEASTER'S
ANTHERCULTURE

Conditions for haploids obtaining at cultivation of anthers of narrow-
leaved oleaster (Elaeagnus angustifolia L.) have been studied.
Anthers, containing mononucleate microspores at the early and middle
stage of development, were cultivated on different media. It was noted that
androgenesis induction only after callus formation took place. After callus
tissue obtaining two further morphogenetic directions may be observed:
organogenesis and embryogenesis.
Callus formation was noted on the all media used, but the best results
were observed on MS medium with 6-BAP, IBA and 2,4-D added.
Callus replantation on the medium for plant regeneration (MS with 6-
BAP 0,4-0,8 mg/l) promoted the meristem zones formation and a large
quantity of microbuds initiation. Some of microbuds were developed into
normal sprouts.
Formation of numerous embryoids along with the microbuds initiation
was observed. Experiments show that it would be betterto use 6-BAP as a
cytokinin active substance, but IBA and 2,4-D as auxin active ones fo rthe
embryogenetic structures development. The total amount of hormones should
not exceed 0,5 mg/l. The embryoids obtained were able to transform into
normal regenerants with a well-developed root system after moving them on
the hormoneless medium or on the medium with minimum hormones content
(up to 0,1 m g /l).
